Recombinant Human Proliferating Cell Nuclear Antigen

  • Description: Recombinant Human Proliferating Cell Nuclear Antigen
  • Product Name Alternative: PCNA
  • CAS Number: 9000-83-3
  • Gene Name: PCNA
  • UniProt: P12004
  • Cellular Locus: Nucleus , Note=Colocalizes with CREBBP, EP300 and POLD1 to sites of DNA damage (PubMed:24939902). Forms nuclear foci representing sites of ongoing DNA replication and vary in morphology and number during S phase (PubMed:15543136). Co-localizes with SMARCA5/SNF2H and BAZ1B/WSTF at replication foci during S phase (PubMed:15543136). Together with APEX2, is redistributed in discrete nuclear foci in presence of oxidative DNA damaging agents.
  • Reactivity: Human
  • Target: Proliferating Cell Nuclear Antigen
  • Type: Protein
  • Applications: WB
  • Concentration: Lot Specific
  • Dilution: Dilute in PBS or medium that is identical to that used in the assay system.
  • Form: Liquid
  • Buffer: 20 mM Tris
  • Function: Auxiliary protein of DNA polymerase delta and is involved in the control of eukaryotic DNA replication by increasing the polymerase's processibility during elongation of the leading strand. Induces a robust stimulatory effect on the 3'-5' exonuclease and 3'-phosphodiesterase, but not apurinic-apyrimidinic (AP) endonuclease, APEX2 activities. Has to be loaded onto DNA in order to be able to stimulate APEX2. Plays a key role in DNA damage response (DDR) by being conveniently positioned at the replication fork to coordinate DNA replication with DNA repair and DNA damage tolerance pathways (PubMed:24939902). Acts as a loading platform to recruit DDR proteins that allow completion of DNA replication after DNA damage and promote postreplication repair: Monoubiquitinated PCNA leads to recruitment of translesion (TLS) polymerases, while 'Lys-63'-linked polyubiquitination of PCNA is involved in error-free pathway and employs rPubMed:18719106, PubMed:19443450, PubMed:24695737, PubMed:24939902}.
  • Storage Conditions: Although stable at 4°C for 4 weeks, product is best stored at or below -20°C. Addition of a carrier protein (such as 0.1% HSA or BSA) is recommended for long-term storage. For in vitro investigational use only.
  • Specificity: Proliferating Cell Nuclear Antigen
  • Formulation: Sterile-filtered solution in 20mM Tris, pH 7.5, 2mM EDTA, and 20% glycerol.
  • Buffer pH: pH 7.5
  • Target Background: PCNA is located in the nucleus and is a cofactor of DNA polymerase delta. PCNA acts as a homotrimer and helps in processing of leading strand synthesis during DNA replication. In reaction to DNA damage, PCNA is ubiquitinated and participates in the RAD6-dependent DNA repair pathway. Two transcript variants encoding the same protein have been found for PCNA gene. PCNA is expressed during late G1 phase, S phase of mitosis, and persists until the end of the M phase. PCNA can be induced by UV irradiation.
