  • FPC or Phycocyanin is a phycobiliprotein isolated from Spirulina sp., a blue-green algae. Like other phycobiliproteins, C-Phycocyanin is fluorescent, with an extremely high absorbtivity, a high quantum efficiency, a large Stokes shift and excitation and emission bands at visible wavelengths. It is a stable protein which can be easily linked to antibodies and other proteins by conventional protein cross-linking techniques without altering its spectral characteristics.

 

  • PE or Phycoerythrin is a phycobiliprotein isolated from red algae developed as a source of choice because it yields one of the most highly fluorescent. Like other phycobiliproteins, R-PE is fluorescent, with an extremely high absorbtivity, a high quantum efficiency, a large Stokes shift and excitation and emission bands at visible wavelengths. It is a stable protein which can be easily linked to antibodies and other proteins by conventional protein cross-linking techniques without altering its spectral characteristics.

 

To be used in fluorescence analysis, FPC or PE has the unique advantages compared with traditional chemic fluorescent dye, they main lie below:

1) It possesses wider absorption spectrum when it is in the wider range of PH, and it is easier to select appropriate excitation wavelength in order to take fluorescence emission with efficiency, in addition, it has differential fluorescence-emission peak;

2) Absorbance and fluorescence quantum productivity is very high, the fluorescence is strong and stable, the sensitivity is high, too;

3) It has smaller fluorescence background and the fluorescence emission is not easy to quench, its fluorescence retention period is longer;

4) Its water solubility is excellent, and its prone to cross link other molecule(s), and the nonspecific adsorption is small;

5) It is purified from pure natural halobios and no toxicity and side effect, no radioactivity. And the operation is very safe.

 

Because of their properties, phycobiliproteins have been used in a variety of immunological assays and as fluorescent labels for cell-sorting. In addition, because of the high molar absorbtivity of these proteins at visible wavelengths, they are convenient markers in such applications as gel electrophoresis, isoelectric focusing and gel exclusion chromatography.
