One unit is defined as the amount of caspase needed to generate 1 pmole of free amc at 30 degrees C, using DEVD-amc (50 micromolar) as substrate.

Activity was assayed as follows:

Reaction buffer:

Cell-free system buffer: 220 mM mannitol, 68 mM sucrose, 2 mM NaCl, 2.5 mM KH₂PO₄, 2 mM MgCl₂ 10 mM Hepes pH 7.4, 10 mM DTT, 1 mM aprotinin, 1 mM leupeptin and 100 mM phenylmethylsulfonyl fluoride (PMSF). Reaction volumes were 100 microl and the final substrate concentration was 50 microM (DEVD-amc).

Analysis for caspase activity was performed at 37°C in a fluorometer (CytoFluor; PerSeptive Biosystems, Framingham, MA, USA) using an excitation wavelength of 360 nm and an emission wavelength of 460 nm. Monitoring was with 2 minutes interval for a total assay time of 30 minutes. A dilution series of free AMC was used as a standard.
