Instructions for Storage and Use

Product Description
Human Melanocyte Growth Supplement (HM-GMS) is a sterile, concentrated (30X) solution intended for use in a complete culture environment for the growth of human epidermal melanocytes. Each 15 ml bottle of HM-GMS is the correct amount of supplement for a 500 ml bottle of basal HAM F10 nutrient Mixture 500 ml (in a 500ml bottle) to which has to be added Antibiotics, HEPES 6mM, Foetal Bovine Serum (8%) decomplemented at 55°C for 25 min.

Intended Use
HM-GMS is intended for use in conjunction with HMF-10 media for the routine culture of normal human epidermal melanocytes. Additional applications may include primary isolation of melanocytes from skin.

Storage and Stability
HM-GMS is stored at -25° C at our facility and is shipped on dry ice. If the product is to be used immediately, thaw in a 37° C water bath or overnight at 5° C. If thawed in a water bath, do not leave the product at 37° C for an extended period of time. When stored at 5° C, HM-GMS is stable for 1 month. If the product is not to be used within 1 month after receipt, we recommend storing it at -25° C in a freezer that is not self-defrosting. Do not thaw and refreeze more than once. When stored at -25°C, the product is stable until the expiration date shown on the label.

Preparation of Supplemented Culture Medium

For routine culture of human epidermal melanocytes, add one bottle (25 ml) of HM-GMS per 500 ml of an appropriate basal medium.

·        HAM F10 nutrient Mixture 500 ml (in a 500ml bottle)

·        Antibiotics

·        HEPES 6mM

·        Foetal Bovine Serum (8%) decomplemented at 56°C for 25 min.

·        The content of 1 vial of #HM-GMS

Keep the bottle at 5°C until used.  

In case of slow growth rate

It is possible to add 35 nM TPA (phorbol 12-myristate 13-acetate) and 2nM cholera toxin to the culture medium. * List Laboratories 95 Euro/ 5mg

To further accelerate cell growth, one may add to the same culture medium 40µg/ml bovine pituitary extract and 3 ng/ml EGF.
